Na v\u00fdzkumu spolupracovali badatel\u00e9 z \u00dastavu experiment\u00e1ln\u00ed botaniky Akademie v\u011bd \u010cR, z P\u0159\u00edrodov\u011bdeck\u00e9 fakulty Univerzity Karlovy a z n\u011bkolika belgick\u00fdch instituc\u00ed. Objev publikovan\u00fd v \u010dasopise Nature Plants m\u016f\u017ee v budoucnu pomoci p\u0159i p\u0159esn\u011bj\u0161\u00edm \u0159\u00edzen\u00ed bun\u011b\u010dn\u00e9ho d\u011blen\u00ed rostlin i \u0161lecht\u011bn\u00ed zem\u011bd\u011blsk\u00fdch plodin.<\/h6>\n<p>Rostlinn\u00e9 bu\u0148ky funguj\u00ed podobn\u011b jako mal\u00e9 logistick\u00e9 centrum \u2013 neust\u00e1le p\u0159ij\u00edmaj\u00ed a t\u0159\u00edd\u00ed l\u00e1tky pot\u0159ebn\u00e9 pro r\u016fst a komunikaci. Endocyt\u00f3za je proces, p\u0159i n\u011bm\u017e se z povrchu bu\u0148ky odd\u011bluj\u00ed drobn\u00e9 v\u00e1\u010dky nesouc\u00ed r\u016fzn\u00e9 l\u00e1tky dovnit\u0159. Tento mechanismus je z\u00e1sadn\u00ed pro p\u0159\u00edjem n\u011bkter\u00fdch \u017eivin a hormon\u016f, regulaci bun\u011b\u010dn\u00e9ho d\u011blen\u00ed i udr\u017een\u00ed spr\u00e1vn\u00e9 funkce membr\u00e1ny. Prvn\u00ed f\u00e1ze endocyt\u00f3zy, kterou t\u00fdm nyn\u00ed detailn\u011b popsal, je zak\u0159iven\u00ed \u2013 vchl\u00edpen\u00ed \u2013 mal\u00e9 oblasti povrchov\u00e9 membr\u00e1ny sm\u011brem dovnit\u0159 bu\u0148ky. Z membr\u00e1ny se v tomto m\u00edst\u011b nakonec zformuje mikroskopick\u00fd v\u00e1\u010dek, kter\u00fd se odd\u011bl\u00ed a putuje se sv\u00fdm \u201en\u00e1kladem\u201c na m\u00edsto ur\u010den\u00ed.<\/p>\n<h6>Jak bu\u0148ka startuje svou dopravu<\/h6>\n<p>Belgicko-\u010desk\u00fd t\u00fdm veden\u00fd Dani\u00eblem Van Dammem z institutu VIB Center for Plant Systems Biology a Univerzity v Gentu a Romanem Pleskotem z \u00dastavu experiment\u00e1ln\u00ed botaniky Akademie v\u011bd \u010cR (\u00daEB AV \u010cR) spojil experimenty a v\u00fdpo\u010detn\u00ed simulace a zkoumal, jak proteinov\u00fd komplex TPLATE spou\u0161t\u00ed prvn\u00ed f\u00e1zi endocyt\u00f3zy. <em>\u201eU\u017e p\u0159ed \u010dasem jsme dok\u00e1zali, \u017ee komplex je nutn\u00fd pro zak\u0159iven\u00ed membr\u00e1ny na povrchu bu\u0148ky. Nyn\u00ed jsme zjistili, jak to pravd\u011bpodobn\u011b d\u011bl\u00e1,\u201c<\/em> vysv\u011btluje Roman Pleskot z \u00daEB AV \u010cR. \u201e<em>Nejprve jsme vytvo\u0159ili podrobn\u00fd model struktury komplexu TPLATE. Struktura n\u00e1m poskytuje mnoho informac\u00ed o funkci molekul \u2013 m\u016f\u017eeme to p\u0159irovnat k d\u016fkladn\u00e9mu prohl\u00e9dnut\u00ed j\u00edzdn\u00edho kola, kdy dok\u00e1\u017eete snadno pochopit, jak funguje a k \u010demu slou\u017e\u00ed,\u201c<\/em> dod\u00e1v\u00e1.<\/p>\n<p>Uk\u00e1zalo se, \u017ee komplex TPLATE je tvo\u0159en dv\u011bma odli\u0161n\u00fdmi \u010d\u00e1stmi. Zjednodu\u0161en\u011b \u0159e\u010deno, jedna \u010d\u00e1st m\u00e1 pevnou strukturu, zat\u00edmco druh\u00e1 m\u016f\u017ee flexibiln\u011b m\u011bnit tvar. Matematick\u00e9 simulace molekul\u00e1rn\u00edch interakc\u00ed mezi komplexem a membr\u00e1nou potvrdily, \u017ee endocyt\u00f3za za\u010d\u00edn\u00e1 tehdy, kdy\u017e se tv\u00e1rn\u00e9 \u010d\u00e1sti alespo\u0148 dvou komplex\u016f setkaj\u00ed na povrchu membr\u00e1ny. Teprve jejich vz\u00e1jemn\u00e1 spolupr\u00e1ce s pevn\u011bj\u0161\u00edmi \u010d\u00e1stmi umo\u017en\u00ed vznik charakteristick\u00e9 vchl\u00edpeniny, z n\u00ed\u017e se n\u00e1sledn\u011b odd\u011bl\u00ed transportn\u00ed v\u00e1\u010dek.<\/p>\n<p><em>\u201eSimulace d\u011bj\u016f na molekul\u00e1rn\u00ed \u00farovni n\u00e1m slou\u017e\u00ed jako \u201av\u00fdpo\u010detn\u00ed mikroskop\u2018. D\u00edky n\u011bmu vid\u00edme v\u00edce, ne\u017e bychom vid\u011bli sebelep\u0161\u00edm fyzick\u00fdm mikroskopem. Podstatn\u00e1 je tak\u00e9 souhra mezi experiment\u00e1ln\u00edm a v\u00fdpo\u010detn\u00edm p\u0159\u00edstupem k probl\u00e9mu. V\u00fdsledky pokus\u016f m\u016f\u017eeme ov\u011b\u0159ovat simulacemi a naopak, co\u017e zefektiv\u0148uje pr\u00e1ci a umo\u017e\u0148uje \u0159e\u0161it i velmi slo\u017eit\u00e9 ot\u00e1zky,\u201c<\/em> \u0159\u00edk\u00e1 spoluautorka \u010dl\u00e1nku Michaela Neubergerov\u00e1, doktorsk\u00e1 studentka P\u0159\u00edrodov\u011bdeck\u00e9 fakulty Univerzity Karlovy, kter\u00e1 z\u00e1rove\u0148 pracuje v laborato\u0159i Romana Pleskota v \u00daEB AV \u010cR.<\/p>\n<h6>Dopady pro v\u00fdzkum a evoluci<\/h6>\n<p>Roman Pleskot z\u00edskal v roce 2022 presti\u017en\u00ed p\u011btilet\u00fd grant Junior STAR ur\u010den\u00fd mlad\u00fdm v\u011bdc\u016fm pro vybudov\u00e1n\u00ed vlastn\u00ed v\u00fdzkumn\u00e9 skupiny. Hlavn\u00edm t\u00e9matem jeho t\u00fdmu jsou nyn\u00ed procesy zapojen\u00e9 do d\u011blen\u00ed rostlinn\u00fdch bun\u011bk \u2013 v\u010detn\u011b endocyt\u00f3zy. \u201e<em>Kdy\u017e detailn\u011b porozum\u00edme molekul\u00e1rn\u00edm mechanism\u016fm, budeme moci bun\u011b\u010dn\u00e9 d\u011blen\u00ed p\u0159esn\u011b regulovat. To je d\u016fle\u017eit\u00e9 i pro \u0161lecht\u011bn\u00ed lep\u0161\u00edch odr\u016fd plodin. V\u00fdsledky projektu otev\u00edraj\u00ed r\u016fzn\u00e9 mo\u017enosti dal\u0161\u00edho v\u00fdzkumu \u2013 m\u016f\u017eeme zkoumat dal\u0161\u00ed f\u00e1ze endocyt\u00f3zy a jej\u00ed evoluci,\u201c<\/em> dod\u00e1v\u00e1 Roman Pleskot.<\/p>\n<p>Komplex TPLATE je evolu\u010dn\u011b star\u00e9 \u0159e\u0161en\u00ed, kter\u00e9 chyb\u00ed u \u017eivo\u010dich\u016f a kvasinek, kde endocyt\u00f3za prob\u00edh\u00e1 odli\u0161n\u011b.
