D\u00edky t\u00e9to inovaci poprv\u00e9 p\u0159\u00edmo ovlivnili d\u011bn\u00ed uvnit\u0159 bun\u011bk a zjistili, \u017ee pro-rakovinn\u00e9 v\u00e1\u010dky s proteinem Rab25 hraj\u00ed kl\u00ed\u010dovou roli p\u0159i tvorb\u011b v\u00fdb\u011b\u017ek\u016f, je\u017e umo\u017e\u0148uj\u00ed n\u00e1dorov\u00fdm bu\u0148k\u00e1m pronikat do okoln\u00ed tk\u00e1n\u011b. Tento objev, publikovan\u00fd v presti\u017en\u00edm \u010dasopise Science Advances, otev\u00edr\u00e1 nov\u00e9 mo\u017enosti pro pochopen\u00ed a potenci\u00e1ln\u00ed ovlivn\u011bn\u00ed \u0161\u00ed\u0159en\u00ed rakoviny v t\u011ble.<\/h6>\n<p>A\u017e dosud v\u011bdci tu\u0161ili, \u017ee recykla\u010dn\u00ed v\u00e1\u010dky neboli endozomy hraj\u00ed d\u016fle\u017eitou roli v pohybu a chov\u00e1n\u00ed bun\u011bk, ale nedok\u00e1zali p\u0159\u00edmo zjistit, jak ovliv\u0148uj\u00ed tvorbu invazivn\u00edch v\u00fdb\u011b\u017ek\u016f rakovinn\u00fdch bun\u011bk. T\u00fdmu Jakuba Gemperleho z \u00dastavu molekul\u00e1rn\u00ed genetiky AV \u010cR (\u00daMG AV \u010cR) se to nyn\u00ed poda\u0159ilo ve spolupr\u00e1ci s britsk\u00fdmi, n\u011bmeck\u00fdmi a francouzsk\u00fdmi kolegy. Pou\u017eili inovativn\u00ed metodu, kter\u00e1 kombinuje molekul\u00e1rn\u00ed biologii s magnetickou manipulac\u00ed \u2013 jmenuje se magnetogenetika.<\/p>\n<p>Pomoc\u00ed magnetick\u00fdch nano\u010d\u00e1stic a p\u0159esn\u011b zac\u00edlen\u00e9ho magnetick\u00e9ho pole doslova \u201ep\u0159it\u00e1hli\u201c endozomy s proteinem Rab25 bl\u00ed\u017ee k okraji \u017eiv\u00fdch rakovinn\u00fdch bun\u011bk. Bu\u0148ky okam\u017eit\u011b za\u010daly tvo\u0159it mikroskopick\u00e9 v\u00fdb\u011b\u017eky, kter\u00e9 jim pom\u00e1haj\u00ed pronikat do okoln\u00ed tk\u00e1n\u011b. Jde o jasn\u00fd d\u016fkaz, \u017ee pouh\u00e1 p\u0159\u00edtomnost t\u011bchto endozom\u016f na okraji bun\u011bk spou\u0161t\u00ed agresivn\u00ed chov\u00e1n\u00ed rakoviny.<\/p>\n<h6>Rab25 jako dirigent bun\u011b\u010dn\u00e9 invaze<\/h6>\n<p>Protein Rab25, o n\u011bm\u017e je zn\u00e1mo, \u017ee souvis\u00ed s agresivn\u00edmi typy rakoviny prsu a vaje\u010dn\u00edk\u016f, se v tomto v\u00fdzkumu prok\u00e1zal jako hlavn\u00ed dirigent cel\u00e9ho procesu. Nejen\u017ee p\u0159en\u00e1\u0161\u00ed d\u016fle\u017eit\u00e9 molekuly (jako je nap\u0159\u00edklad vazebn\u00fd receptor integrin \u03b21 nebo regul\u00e1tor bun\u011b\u010dn\u00e9 kostry FMNL1), ale tak\u00e9 propojuje jejich doru\u010den\u00ed s aktivac\u00ed sign\u00e1l\u016f p\u0159\u00edmo na bun\u011b\u010dn\u00e9 membr\u00e1n\u011b. V\u00fdsledkem je vznik speci\u00e1ln\u00edch v\u00fdb\u011b\u017ek\u016f, kter\u00fdmi rakovinn\u00e9 bu\u0148ky \u201eosah\u00e1vaj\u00ed\u201c sv\u00e9 okol\u00ed a pak se do n\u011bj za\u010dnou aktivn\u011b zapl\u00e9tat a pronikat. Tak si rakovina raz\u00ed cestu d\u00e1l t\u011blem.<\/p>\n<p><em>\u201eNa\u0161\u00edm c\u00edlem bylo pomoc\u00ed magnet\u016f p\u0159esn\u011b zac\u00edlit a ovl\u00e1dat pohyb drobn\u00fdch \u201arecykla\u010dn\u00edch v\u00e1\u010dk\u016f\u2018 uvnit\u0159 rakovinn\u00fdch bun\u011bk. D\u00edky tomu jsme cht\u011bli l\u00e9pe pochopit, jak se rakovina \u0161\u00ed\u0159\u00ed, a naj\u00edt nov\u00e9 zp\u016fsoby, jak jej\u00ed invazi zastavit,\u201c<\/em> \u0159\u00edk\u00e1 Jakub Gemperle, vedouc\u00ed v\u00fdzkumu a v\u011bdeck\u00fd pracovn\u00edk v Laborato\u0159i integrativn\u00ed biologie \u00daMG AV \u010cR.<\/p>\n<p>Z\u00e1sadn\u00edm p\u0159\u00ednosem studie je skute\u010dnost, \u017ee v\u011bdci byli schopni tyto zhoubn\u00e9 mechanismy nejen pozorovat, ale i um\u011ble spou\u0161t\u011bt. V\u00fdznamn\u011b tak pos\u00edlili hypot\u00e9zu, \u017ee Rab25 nen\u00ed pouze pasivn\u00edm \u00fa\u010dastn\u00edkem, ale aktivn\u00edm inici\u00e1torem invazivn\u00ed migrace.<\/p>\n<h6>Nad\u011bje pro c\u00edlenou terapii a lep\u0161\u00ed pochopen\u00ed nejen rakovinotvorn\u00fdch proces\u016f<\/h6>\n<p>V\u00fdsledky v\u00fdzkumu poskytuj\u00ed nov\u00fd \u00fahel pohledu na proces metastazov\u00e1n\u00ed a jeho mo\u017en\u00e9 naru\u0161en\u00ed. Pokud toti\u017e protein Rab25 hraje tak p\u0159\u00edmou roli ve spou\u0161t\u011bn\u00ed invaze, jeho c\u00edlen\u00e9 ovlivn\u011bn\u00ed \u2013 nap\u0159\u00edklad pomoc\u00ed specifick\u00fdch inhibitor\u016f (l\u00e1tek, kter\u00e9 zpomaluj\u00ed nebo zcela zastavuj\u00ed ur\u010ditou chemickou reakci \u010di proces) nebo naru\u0161en\u00edm jeho lokalizace \u2013 by mohlo v\u00fdznamn\u011b zpomalit nebo zastavit \u0161\u00ed\u0159en\u00ed rakoviny v t\u011ble.<\/p>\n<p>Experimenty byly provedeny nejen v klasick\u00fdch bun\u011b\u010dn\u00fdch kultur\u00e1ch, ale i v trojrozm\u011brn\u00e9m prost\u0159ed\u00ed simuluj\u00edc\u00edm p\u0159irozen\u00e9 podm\u00ednky v t\u011ble. Pr\u00e1v\u011b v t\u011bchto slo\u017eit\u011bj\u0161\u00edch podm\u00ednk\u00e1ch se uk\u00e1zalo, \u017ee manipulace s Rab25 vezikuly, tj. mal\u00fdmi, membr\u00e1nou obalen\u00fdmi v\u00e1\u010dky v bu\u0148k\u00e1ch, kter\u00e9 slou\u017e\u00ed k transportu a skladov\u00e1n\u00ed l\u00e1tek, z\u00e1sadn\u011b ovliv\u0148uje schopnost rakovinn\u00fdch bun\u011bk migrovat a kolonizovat nov\u00e9 oblasti.<\/p>\n<p><em>\u201eTento objev m\u011bn\u00ed pohled na to, jak to v bu\u0148k\u00e1ch funguje. D\u0159\u00edve se myslelo, \u017ee endozomy pouze usnad\u0148uj\u00ed um\u00edst\u011bn\u00ed n\u00e1kladu pro vytv\u00e1\u0159en\u00ed v\u00fdb\u011b\u017ek\u016f p\u0159i pohybu bun\u011bk. My jsme ale uk\u00e1zali, \u017ee maj\u00ed p\u0159\u00edmou a z\u00e1sadn\u00ed roli \u2013 jsou lok\u00e1ln\u00edm spou\u0161t\u011b\u010dem.
