To je podstata nov\u00e9ho syst\u00e9mu, kter\u00fd ve studii publikovan\u00e9 v \u010dasopise Journal of Controlled Release p\u0159edstavil t\u00fdm Mikrobiologick\u00e9ho \u00fastavu AV \u010cR a \u00dastavu makromolekul\u00e1rn\u00ed chemie AV \u010cR ve spolupr\u00e1ci s l\u00e9ka\u0159i z Fakultn\u00ed nemocnice v Motole. V\u011bdci a v\u011bdkyn\u011b koncept \u00fasp\u011b\u0161n\u011b otestovali na bu\u0148k\u00e1ch i zv\u00ed\u0159ec\u00edch modelech. M\u00e1 proto potenci\u00e1l st\u00e1t se sou\u010d\u00e1st\u00ed modern\u00ed onkologick\u00e9 l\u00e9\u010dby.<\/h6>\n<p>Chemoterapie je spolu s chirurgick\u00fdm z\u00e1krokem, radioterapi\u00ed a imunoterapi\u00ed jedn\u00edm ze z\u00e1kladn\u00edch zp\u016fsob\u016f l\u00e9\u010dby rakoviny. Velk\u00fdm probl\u00e9mem je v\u0161ak tzv. mnoho\u010detn\u00e1 l\u00e9kov\u00e1 rezistence. Jedn\u00edm z jej\u00edch \u010dast\u00fdch mechanism\u016f je \u201epumpov\u00e1n\u00ed\u201c l\u00e9\u010diva z bu\u0148ky d\u0159\u00edv, ne\u017e za\u010dne p\u016fsobit, pomoc\u00ed zvl\u00e1\u0161tn\u00edch transportn\u00edch protein\u016f. V membr\u00e1n\u011b \u0159ady typ\u016f zdrav\u00fdch bun\u011bk se vyskytuj\u00ed b\u011b\u017en\u011b a umo\u017e\u0148uj\u00ed p\u0159enos cel\u00e9 \u0159ady l\u00e1tek p\u0159es membr\u00e1nu bu\u0148ky. U rezistentn\u00edch rakovinn\u00fdch bun\u011bk se ale n\u011bkter\u00e9 transportn\u00ed proteiny v extr\u00e9mn\u00edm po\u010dtu zmno\u017e\u00ed a p\u016fsoben\u00ed l\u00e9\u010diva uvnit\u0159 bun\u011bk zabr\u00e1n\u00ed, proto\u017ee jsou schopn\u00e9 l\u00e9\u010divo \u00fapln\u011b nebo t\u00e9m\u011b\u0159 \u00fapln\u011b z bu\u0148ky vylou\u010dit.<\/p>\n<p><em>\u201eZam\u011b\u0159ili jsme se na rezistentn\u00ed rakovinn\u00e9 bu\u0148ky, kter\u00e9 maj\u00ed hodn\u011b tzv. P-glykoprotein\u016f,\u201c<\/em> \u0159\u00edk\u00e1 Milada \u0160\u00edrov\u00e1 z Laborato\u0159e n\u00e1dorov\u00e9 imunologie Mikrobiologick\u00e9ho \u00fastavu AV \u010cR. Nov\u00e9 l\u00e1tky jej\u00ed t\u00fdm testoval in vitro (ve zkumavce) a tak\u00e9 in vivo \u2013 na my\u0161\u00edch. \u201e<em>D\u00edky koleg\u016fm z Fakultn\u00ed nemocnice Motol jsme nav\u00edc mohli pou\u017e\u00edt speci\u00e1ln\u00ed my\u0161\u00ed model s n\u00e1dorem klinick\u00e9ho p\u016fvodu. Vzorky odebran\u00e9 z n\u00e1dor\u016f pacient\u016f toti\u017e mohou za ur\u010dit\u00fdch okolnost\u00ed r\u016fst v experiment\u00e1ln\u00edch my\u0161\u00edch a lze na nich otestovat \u00fa\u010dinek l\u00e9\u010diva. Mo\u017enost takto pracovat se vzorky n\u00e1dor\u016f pacient\u016f p\u0159in\u00e1\u0161\u00ed dal\u0161\u00ed kvalitu p\u0159i testov\u00e1n\u00ed: tento syst\u00e9m je o n\u011bco bl\u00ed\u017ee skute\u010dn\u00fdm podm\u00ednk\u00e1m p\u0159i l\u00e9\u010db\u011b. To je velmi d\u016fle\u017eit\u00e9 pro zhodnocen\u00ed potenci\u00e1lu nov\u00e9 l\u00e1tky pro klinick\u00e9 vyu\u017eit\u00ed,\u201c<\/em> zd\u016fraz\u0148uje Milada \u0160\u00edrov\u00e1.<\/p>\n<h6>C\u00edlen\u00fd syst\u00e9m dopravy l\u00e9\u010div v t\u011ble<\/h6>\n<p>Z\u00e1sadn\u00edm p\u0159\u00ednosem byla spolupr\u00e1ce biolog\u016f \u2013 imunolog\u016f s chemiky, kte\u0159\u00ed navrhli, p\u0159ipravili a charakterizovali dopravn\u00ed syst\u00e9m pro l\u00e9\u010divo na b\u00e1zi syntetick\u00fdch kopolymer\u016f. Tyto speci\u00e1ln\u00ed amfifiln\u00ed kopolymery (PHPMA-b-PPO) se ve vodn\u00e9m prost\u0159ed\u00ed samy skl\u00e1daj\u00ed do micel \u2013 mal\u00fdch kulovit\u00fdch struktur schopn\u00fdch pasivn\u011b zac\u00edlit l\u00e9\u010divo p\u0159\u00edmo do oblasti n\u00e1dorov\u00e9 tk\u00e1n\u011b. \u201e<em>Do t\u011bchto struktur jsme nav\u00e1zali doxorubicin \u2013 b\u011b\u017en\u00e9 cytostatikum pou\u017e\u00edvan\u00e9 k l\u00e9\u010db\u011b rakoviny. Kopolymer jej dok\u00e1\u017ee p\u0159esn\u011b nasm\u011brovat do n\u00e1doru. To je kl\u00ed\u010dov\u00e9 pro to, aby byl l\u00e9k \u00fa\u010dinn\u00fd a vedlej\u0161\u00ed \u00fa\u010dinky l\u00e9\u010dby co nejm\u00e9n\u011b zat\u00ed\u017eily zdrav\u00e9 tk\u00e1n\u011b pacienta,\u201c<\/em> vysv\u011btluje Tom\u00e1\u0161 Etrych, vedouc\u00ed Odd\u011blen\u00ed biol\u00e9ka\u0159sk\u00fdch polymer\u016f \u00dastavu makromolekul\u00e1rn\u00ed chemie AV \u010cR.<\/p>\n<h6>Omezen\u00ed zdroje energie v rakovinn\u00e9 bu\u0148ce<\/h6>\n<p>K vylu\u010dov\u00e1n\u00ed ne\u017e\u00e1douc\u00edch l\u00e1tek z bun\u011bk (toxiny, metabolity, l\u00e9\u010diva) vyu\u017e\u00edvaj\u00ed bu\u0148ky energii ve form\u011b ATP. ATP je kl\u00ed\u010dov\u00e1 molekula pot\u0159ebn\u00e1 pro fungov\u00e1n\u00ed v\u011bt\u0161iny bun\u011b\u010dn\u00fdch proces\u016f. Jej\u00edm zdrojem jsou mitochondrie, je\u017e zaji\u0161\u0165uj\u00ed krom\u011b tvorby ATP i bun\u011b\u010dn\u00e9 d\u00fdch\u00e1n\u00ed.
