Srde\u010dn\u00ed bu\u0148ky v takov\u00e9m prost\u0159ed\u00ed aktivuj\u00ed proteiny, jako je nap\u0159\u00edklad HIF-1\u03b1, kter\u00e9 nastartuj\u00ed molekul\u00e1rn\u00ed mechanismy k obran\u011b. T\u00fdm z Fyziologick\u00e9ho \u00fastavu Akademie v\u011bd \u010cR zjistil, \u017ee p\u0159\u00edtomnost proteinu HIF-1\u03b1 je kl\u00ed\u010dov\u00e1 pro aktivn\u00ed odstra\u0148ov\u00e1n\u00ed nefunk\u010dn\u00edch \u010di nadbyte\u010dn\u00fdch mitochondri\u00ed, tj. bun\u011b\u010dn\u00fdch tov\u00e1ren na energii. Odhalil tak nov\u00fd bun\u011b\u010dn\u00fd mechanismus, kter\u00fd pom\u00e1h\u00e1 ochr\u00e1nit srdce p\u0159ed d\u016fsledky akutn\u00edho infarktu myokardu.<\/h6>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"size-medium wp-image-243085 alignright\" src=\"https:\/\/zakrasnejsivimperk.cz\/wp-content\/uploads\/2021\/12\/19\/akademie-v\u011bd-av-cr-logo-300x80.png\" alt=\"\" width=\"300\" height=\"80\" srcset=\"https:\/\/zakrasnejsivimperk.cz\/wp-content\/uploads\/2021\/12\/19\/akademie-v\u011bd-av-cr-logo-300x80.png 300w, https:\/\/zakrasnejsivimperk.cz\/wp-content\/uploads\/2021\/12\/19\/akademie-v\u011bd-av-cr-logo-768x206.png 768w, https:\/\/zakrasnejsivimperk.cz\/wp-content\/uploads\/2021\/12\/19\/akademie-v\u011bd-av-cr-logo-1024x275.png 1024w, https:\/\/zakrasnejsivimperk.cz\/wp-content\/uploads\/2021\/12\/19\/akademie-v\u011bd-av-cr-logo-1331x357.png 1331w, https:\/\/zakrasnejsivimperk.cz\/wp-content\/uploads\/2021\/12\/19\/akademie-v\u011bd-av-cr-logo-1801x483.png 1801w\" sizes=\"(max-width: 300px) 100vw, 300px\" \/>Aby v\u011bdci tento mechanismus prozkoumali, provedli pod z\u00e1\u0161titou N\u00e1rodn\u00edho institutu pro v\u00fdzkum metabolick\u00fdch a kardiovaskul\u00e1rn\u00edch onemocn\u011bn\u00ed studii s dv\u011bma skupinami my\u0161\u00ed \u2013 b\u011b\u017en\u00e9ho typu (wild-type) a se sn\u00ed\u017eenou hladinou proteinu HIF-1\u03b1. N\u011bkter\u00e9 my\u0161i proto byly po dobu \u010dty\u0159 t\u00fddn\u016f vystaveny dlouhodob\u00e9mu nedostatku kysl\u00edku (chronick\u00e9 hypoxii), jin\u00e9 \u017eily ve standardn\u00edch podm\u00ednk\u00e1ch. Srdce obou skupin my\u0161\u00ed byla n\u00e1sledn\u011b vystavena ischemii a reperfuzi napodobuj\u00edc\u00edch proces infarktu myokardu a n\u00e1sledn\u00e9ho obnoven\u00ed krevn\u00edho pr\u016ftoku.<\/p>\n<p><em>\u201eZjistili jsme, \u017ee my\u0161i vystaven\u00e9 n\u00edzk\u00e9mu obsahu kysl\u00edku m\u011bly siln\u011bj\u0161\u00ed srdce, kter\u00e9 l\u00e9pe odolalo po\u0161kozen\u00ed, ne\u017e ty, je\u017e touto adaptac\u00ed nepro\u0161ly. D\u00e1le jsme prok\u00e1zali, \u017ee srde\u010dn\u00ed bu\u0148ky t\u011bchto my\u0161\u00ed vykazovaly z\u00e1sadn\u00ed zm\u011bny. Aktivn\u011b odstra\u0148ovaly po\u0161kozen\u00e9 mitochondrie v procesu zvan\u00e9m mitofagie, aby z\u016fstaly zdrav\u00e9. Tyto zm\u011bny u my\u0161\u00ed se sn\u00ed\u017eenou hladinou proteinu HIF-1\u03b1 nenastaly,\u201c<\/em> popisuje Petra Al\u00e1nov\u00e1, hlavn\u00ed autorka studie z Odd\u011blen\u00ed v\u00fdvojov\u00e9 kardiologie Fyziologick\u00e9ho \u00fastavu AV \u010cR.<\/p>\n<p>T\u00fdm n\u00e1sledn\u011b zji\u0161\u0165oval, zda je tento \u201esamo\u010dist\u00edc\u00ed\u201c proces z\u00e1sadn\u00ed. U n\u011bkter\u00fdch my\u0161\u00ed proto mitofagii zablokovali, a tud\u00ed\u017e na v\u00fdhody nedo\u0161lo a srdce chr\u00e1n\u011bna nebyla. Co\u017e nazna\u010duje, \u017ee proces mitofagie je pro protektivn\u00ed \u00falohu HIF-1\u03b1 kl\u00ed\u010dov\u00fd.<\/p>\n<p><em>\u201eTento v\u00fdzkum n\u00e1m umo\u017e\u0148uje l\u00e9pe porozum\u011bt tomu, jak HIF-1\u03b1 podporuje prosp\u011b\u0161n\u00e9 procesy, kter\u00e9 bu\u0148k\u00e1m umo\u017e\u0148uj\u00ed p\u0159e\u017e\u00edt p\u0159i zv\u00fd\u0161en\u00e9m stresu.
