K tomuto z\u00e1v\u011bru dosp\u011bli v\u011bdci z \u00dastavu \u017eivo\u010di\u0161n\u00e9 fyziologie a genetiky AV \u010cR ve spolupr\u00e1ci s Masarykovou univerzitou a \u00dastavem analytick\u00e9 chemie AV \u010cR. Olovnat\u00e9 nano\u010d\u00e1stice (PbNP), kter\u00e9 se do n\u011bj dost\u00e1vaj\u00ed z pr\u016fmyslov\u00e9 v\u00fdroby a spalov\u00e1n\u00edm fosiln\u00edch paliv, mohou p\u0159edstavovat skryt\u00e9 nebezpe\u010d\u00ed.<\/p>\n<h6>Nano\u010d\u00e1stice olova obch\u00e1zej\u00ed p\u0159irozenou ochranu mozku<\/h6>\n<p>V\u011bdci zjistili, \u017ee tyto \u010d\u00e1stice mohou vstoupit do mozkov\u00e9 tk\u00e1n\u011b p\u0159\u00edmo p\u0159es \u010dichov\u00e9 bu\u0148ky, ani\u017e by musely p\u0159ekon\u00e1vat hematoencefalickou bari\u00e9ru, kter\u00e1 odd\u011bluje vnit\u0159n\u00ed prost\u0159ed\u00ed mozku od c\u00e9vn\u00edho syst\u00e9mu v t\u011ble a br\u00e1n\u00ed tak, aby do n\u011bj vstupovaly \u0161kodliv\u00e9 l\u00e1tky. \u201e<em>Potvrdili jsme, \u017ee olovnat\u00e9 nano\u010d\u00e1stice se mohou dostat do mozku p\u0159es \u010dichov\u00fd nerv a hromadit se ve specifick\u00fdch oblastech mozku,\u201c<\/em> vysv\u011btluje Marcela Buchtov\u00e1, vedouc\u00ed Laborato\u0159e molekul\u00e1rn\u00ed morfogeneze \u00dastavu \u017eivo\u010di\u0161n\u00e9 fyziologie a genetiky (\u00da\u017dFG AV \u010cR). \u201e<em>Tento objev je kl\u00ed\u010dov\u00fd pro pochopen\u00ed neurotoxicity nano\u010d\u00e1stic a ukazuje, \u017ee zne\u010di\u0161t\u011bn\u00ed ovzdu\u0161\u00ed m\u016f\u017ee m\u00edt p\u0159\u00edm\u00fd dopad na n\u00e1\u0161 nervov\u00fd syst\u00e9m. Z\u00e1rove\u0148 otev\u00edr\u00e1 nov\u00e9 mo\u017enosti ochrany na\u0161eho zdrav\u00ed.\u201c<\/em><\/p>\n<h6>Hromad\u011bn\u00ed nano\u010d\u00e1stic v mozku a neurodegenerativn\u00ed zm\u011bny<\/h6>\n<p>Studie prok\u00e1zala, \u017ee zejm\u00e9na olovnat\u00e9 nano\u010d\u00e1stice s n\u00edzkou rozpustnost\u00ed maj\u00ed tendenci se v mozku hromadit a jejich odstran\u011bn\u00ed je velmi obt\u00ed\u017en\u00e9. I po dlouh\u00e9 dob\u011b, kdy mozek nebyl \u010d\u00e1stic\u00edm vystaven, toti\u017e z\u016fst\u00e1valy v mozkov\u00e9 tk\u00e1ni vysok\u00e9 hladiny olova, zejm\u00e9na v hipokampu \u2013 oblasti kl\u00ed\u010dov\u00e9 pro pam\u011b\u0165 a u\u010den\u00ed. To znamen\u00e1, \u017ee dlouhodob\u00e1 expozice m\u016f\u017ee v\u00e9st k v\u00e1\u017en\u00fdm neurologick\u00fdm d\u016fsledk\u016fm. \u201e<em>Nano\u010d\u00e1stice toti\u017e naru\u0161uj\u00ed cytoskelet neuron\u016f a zp\u016fsobuj\u00ed hromad\u011bn\u00ed shluk\u016f vl\u00e1ken F-aktinu (tzv. Hirano t\u011bl\u00edsek), co\u017e je zn\u00e1mka neurodegenerativn\u00edch proces\u016f,\u201c<\/em> vysv\u011btluje Jana Dumkov\u00e1 z Masarykovy univerzity. V\u011bdeck\u00fd t\u00fdm nav\u00edc zjistil poruchu fosforylace tau proteinu \u2013 procesu spojen\u00e9ho s Alzheimerovou chorobou a dal\u0161\u00edmi tauopatiemi. \u201e<em>Vid\u00edme jasnou souvislost mezi expozic\u00ed olovnat\u00fdm nano\u010d\u00e1stic\u00edm a vznikem zm\u011bn typick\u00fdch pro neurodegenerativn\u00ed onemocn\u011bn\u00ed. Doch\u00e1z\u00ed i k po\u0161kozen\u00ed bari\u00e9ry, kter\u00e1 norm\u00e1ln\u011b reguluje transport l\u00e1tek mezi krv\u00ed a mozkem, a t\u00edm d\u00e1le zvy\u0161uje riziko neurotoxick\u00fdch \u00fa\u010dink\u016f t\u011bchto nano\u010d\u00e1stic,\u201c<\/em> dopl\u0148uje Marcela Buchtov\u00e1.<\/p>\n<h6>Olovo jako \u201ene\u017e\u00e1douc\u00ed n\u00e1hra\u017eka\u201c v\u00e1pn\u00edku<\/h6>\n<p>V d\u016fsledku chemick\u00e9 podobnosti m\u016f\u017ee olovo v nervov\u00e9 soustav\u011b napodobovat v\u00e1pn\u00edk, kter\u00fd je kl\u00ed\u010dov\u00fd pro spr\u00e1vn\u00e9 fungov\u00e1n\u00ed mnoha bun\u011b\u010dn\u00fdch proces\u016f, jako je bun\u011b\u010dn\u00e1 signalizace nebo stabilita cytoskeletu. Studie odhalila, \u017ee expozice PbNP zp\u016fsobuje nerovnov\u00e1hu prvk\u016f v mozkov\u00e9 tk\u00e1ni, co\u017e m\u016f\u017ee d\u00e1le negativn\u011b ovlivnit nervovou soustavu. \u201e<em>Na\u0161e experimenty uk\u00e1zaly, \u017ee pokud je sn\u00ed\u017eena hladina v\u00e1pn\u00edku v prost\u0159ed\u00ed, negativn\u00ed \u00fa\u010dinky olovnat\u00fdch nano\u010d\u00e1stic se je\u0161t\u011b zesiluj\u00ed,\u201c<\/em> \u0159\u00edk\u00e1 Daniela Kristekov\u00e1 z \u00dastavu \u017eivo\u010di\u0161n\u00e9 fyziologie a genetiky AV \u010cR, kter\u00e1 studovala vliv nano\u010d\u00e1stic na chov\u00e1n\u00ed modelov\u00e9ho organismu \u2013 ryby zeb\u0159i\u010dky (Danio rerio). U exponovan\u00fdch jedinc\u016f byly pozorov\u00e1ny v\u00fdrazn\u00e9 zm\u011bny v pohybu a koordinaci.<\/p>\n<h6>Mo\u017enosti ochrany nervov\u00e9ho syst\u00e9mu<\/h6>\n<p>V\u011bdci zkoumali i mo\u017enosti, jak negativn\u00ed \u00fa\u010dinky olovnat\u00fdch nano\u010d\u00e1stic na nervovou tk\u00e1\u0148 zm\u00edrnit. <em>\u201eZaj\u00edmav\u00fdm zji\u0161t\u011bn\u00edm je, \u017ee pou\u017eit\u00ed inhibitoru GSK-3, kter\u00fd blokuje fosforylaci tau proteinu, dok\u00e1zalo negativn\u00ed \u00fa\u010dinky PbNP na nervov\u00e9 bu\u0148ky \u010d\u00e1ste\u010dn\u011b zm\u00edrnit,\u201c<\/em> popisuje v\u00fdsledky sv\u00e9 pr\u00e1ce Adriena Jedli\u010dkov\u00e1, kter\u00e1 na projektu za\u010dala pracovat b\u011bhem st\u0159edo\u0161kolsk\u00e9 odborn\u00e9 \u010dinnosti a za sv\u016fj v\u00fdzkum z\u00edskala \u010detn\u00e1 ocen\u011bn\u00ed. Tento poznatek otev\u00edr\u00e1 cestu k dal\u0161\u00edmu v\u00fdzkumu terapeutick\u00fdch strategi\u00ed zam\u011b\u0159en\u00fdch na ochranu mozku p\u0159ed toxick\u00fdmi \u00fa\u010dinky t\u011b\u017ek\u00fdch kov\u016f.<\/p>\n<p>V\u00fdzkum prob\u00edhal v Laborato\u0159i molekul\u00e1rn\u00ed morfogeneze \u00da\u017dFG AV \u010cR ve spolupr\u00e1ci s t\u00fdmy z Masarykovy univerzity (patologick\u00e9 zm\u011bny nervov\u00e9 tk\u00e1n\u011b \u2013 t\u00fdm doc. Ale\u0161e Hampla; anal\u00fdza prvk\u016f v tk\u00e1n\u00edch \u2013 t\u00fdm doc. Tom\u00e1\u0161e Vaculovi\u010de) a \u00dastavu analytick\u00e9 chemie AV \u010cR (inhala\u010dn\u00ed expozice \u2013 t\u00fdm dr. Pavla Miku\u0161ky).
