Biofilmy jsou v\u0161ude kolem n\u00e1s a maj\u00ed z\u00e1sadn\u00ed vliv na \u017eivot na Zemi. Mohou obsahovat bakterie, kvasinky \u010di jin\u00e9 mikroorganismy.<\/p>\n<p>Biofilmy jsou ale v\u00e1\u017en\u00fdm probl\u00e9mem v oborech, jako je medic\u00edna nebo pr\u016fmysl. Kontaminuj\u00ed toti\u017e zdravotnick\u00e9 vybaven\u00ed, nap\u0159\u00edklad kat\u00e9try a implant\u00e1ty, co\u017e m\u016f\u017ee v\u00e9st k chronick\u00fdm infekc\u00edm. V pr\u016fmyslu zase zp\u016fsobuj\u00ed korozi a zne\u010di\u0161t\u011bn\u00ed za\u0159\u00edzen\u00ed. Dokonce i ve vesm\u00edru mohou ohro\u017eovat kl\u00ed\u010dov\u00e9 prvky za\u0159\u00edzen\u00ed jako jednotky na \u00fapravu vody \u010di skafandry.<\/p>\n<h6>Jak p\u0159edej\u00edt tvorb\u011b biofilm\u016f<\/h6>\n<p>Existuj\u00ed dv\u011b hlavn\u00ed strategie, jak se s biofilmy vypo\u0159\u00e1dat. \u201e<em>Prvn\u00ed je ji\u017e vytvo\u0159en\u00e9 biofilmy zlikvidovat. A v\u016fbec nej\u00fa\u010dinn\u011bj\u0161\u00edm zp\u016fsobem, jak se vyhnout probl\u00e9m\u016fm s biofilmy, je zabr\u00e1nit jejich vzniku,<\/em>\u201d \u0159\u00edk\u00e1 vedouc\u00ed v\u00fdzkumn\u00e9ho t\u00fdmu Zdena Palkov\u00e1 z P\u0159\u00edrodov\u011bdeck\u00e9 fakulty Univerzity Karlovy v centru BIOCEV.<\/p>\n<p>Nedovolit mikroorganism\u016fm, aby na povrch p\u0159ilnuly, dok\u00e1\u017eou modern\u00ed metody zahrnuj\u00edc\u00ed antimikrobi\u00e1ln\u00ed \u00fapravy povrch\u016f a nanotechnologie. Pr\u00e1v\u011b na n\u011b se zam\u011b\u0159uje t\u00fdm Zdeny Palkov\u00e9 ve spolupr\u00e1ci s kolegy z laserov\u00e9ho Centra HiLASE v \u010dele s Petrem Hauschwitzem z Fyzik\u00e1ln\u00edho \u00fastavu AV \u010cR.<\/p>\n<p>Spole\u010dn\u00fdm c\u00edlem je navrhnout a zav\u00e9st pokro\u010dil\u00e9 nanostrukturov\u00e9 \u00fapravy povrch\u016f materi\u00e1l\u016f, kter\u00e9 se b\u011b\u017en\u011b pou\u017e\u00edvaj\u00ed v medic\u00edn\u011b a pr\u016fmyslu, t\u00edm v\u00fdrazn\u011b omezit nebo zcela zabr\u00e1nit p\u0159ichyt\u00e1v\u00e1n\u00ed mikrob\u016f, a potla\u010dit tak ne\u017e\u00e1douc\u00ed vznik biofilm\u016f. Mechanick\u00e9 odstran\u011bn\u00ed biofilm\u016f toti\u017e \u010dasto nen\u00ed mo\u017en\u00e9, nap\u0159\u00edklad u implant\u00e1t\u016f v t\u011ble pacienta.<\/p>\n<p><em>\u201eI p\u0159es pokroky ve v\u00fdzkumu st\u00e1le nezn\u00e1me v\u0161echny detaily o biofilmech. Chyb\u011bj\u00ed n\u00e1m informace o jejich vnit\u0159n\u00ed struktu\u0159e a organizaci, co\u017e je kl\u00ed\u010dov\u00e9 pro efektivn\u00ed boj proti nim. Proto je ned\u00edlnou sou\u010d\u00e1st\u00ed na\u0161\u00ed snahy i samotn\u00fd z\u00e1kladn\u00ed v\u00fdzkum, bez kter\u00e9ho nem\u016f\u017eeme odhalit a pochopit mechanismy vztahu mezi r\u016fzn\u011b efektivn\u00ed p\u0159ilnavost\u00ed mikrob\u016f a tvorbou komplexn\u00edch biofilm\u016f,\u201d<\/em> dod\u00e1v\u00e1 Zdena Palkov\u00e1.<\/p>\n<h6>Pokro\u010dil\u00e9 laserov\u00e9 technologie otev\u00edraj\u00ed nov\u00e9 mo\u017enosti<\/h6>\n<p>V\u00fdzkumn\u00fd program RWP3 (Research work package 3; Laser applications in biotechnology) v projektu LasApp se proto zam\u011b\u0159uje na v\u00fdvoj laserov\u00fdch technologi\u00ed pro pokro\u010dil\u00e9 povrchov\u00e9 \u00fapravy vybran\u00fdch materi\u00e1l\u016f ze zdravotnick\u00e9ho a pr\u016fmyslov\u00e9ho odv\u011btv\u00ed, kter\u00e9 jsou \u010dasto pr\u00e1v\u011b biofilmy kontaminov\u00e1ny.<\/p>\n<p><em>\u201eC\u00edlem je vytvo\u0159it povrchy, kter\u00e9 d\u00edky speci\u00e1ln\u00ed struktu\u0159e na mikroskopick\u00e9 a nanometrov\u00e9 \u00farovni br\u00e1n\u00ed p\u0159ilnut\u00ed bakteri\u00ed a dal\u0161\u00edch mikroorganism\u016f,\u201c<\/em> vysv\u011btluje Petr Hauschwitz z Fyzik\u00e1ln\u00edho \u00fastavu AV \u010cR, vedouc\u00ed v\u00fdzkumn\u00e9ho t\u00fdmu laserov\u00e9ho mikroobr\u00e1b\u011bn\u00ed Centra HiLASE.<\/p>\n<p>Laserov\u00e9 strukturov\u00e1n\u00ed je modern\u00ed metoda, kter\u00e1 umo\u017e\u0148uje precizn\u00ed \u00fapravy povrch\u016f materi\u00e1l\u016f na mikro- a nano- \u00farovni. S vyu\u017eit\u00edm \u0161pi\u010dkov\u00fdch laserov\u00fdch technologi\u00ed lze vytv\u00e1\u0159et struktury s detailem v \u0159\u00e1du stovek nanometr\u016f, kter\u00e9 jsou men\u0161\u00ed ne\u017e pr\u016fm\u011brn\u00fd rozm\u011br bakterie nebo mikrobu, co\u017e v\u00fdrazn\u011b sni\u017euje pravd\u011bpodobnost jejich p\u0159ichycen\u00ed na povrch.<\/p>\n<h6>Rychlej\u0161\u00ed a efektivn\u011bj\u0161\u00ed mikro- a nanostruktury pro boj s biofilmy<\/h6>\n<p>Vedle samotn\u00e9ho v\u00fdvoje funk\u010dn\u00ed topografie se t\u00fdmy zam\u011b\u0159uj\u00ed na zefektivn\u011bn\u00ed laserov\u00e9 v\u00fdroby s vyu\u017eit\u00edm pokro\u010dil\u00fdch technik dynamick\u00e9ho tvarov\u00e1n\u00ed laserov\u00e9ho svazku pomoc\u00ed prostorov\u00e9ho sv\u011bteln\u00e9ho modul\u00e1toru (SLM) a interferen\u010dn\u00edho obr\u00e1b\u011bn\u00ed.<\/p>\n<p>Tyto metody umo\u017e\u0148uj\u00ed rychlej\u0161\u00ed a efektivn\u011bj\u0161\u00ed v\u00fdrobu povrch\u016f, kter\u00e9 lze aplikovat i na slo\u017eit\u011b tvarovan\u00e9 p\u0159edm\u011bty. To je velmi d\u016fle\u017eit\u00e9 jak pro pr\u016fmysl, tak pro zdravotnictv\u00ed. Tato inovace m\u016f\u017ee v\u00fdznamn\u011b zlep\u0161it zp\u016fsoby, jak bojovat proti mikrobi\u00e1ln\u00ed kontaminaci.<\/p>\n<p><em>\u201eO\u010dek\u00e1v\u00e1me, \u017ee na\u0161e v\u00fdsledky povedou k v\u00fdvoji nov\u00fdch povrchov\u00fdch \u00faprav, kter\u00e9 budou schopn\u00e9 eliminovat \u0161irok\u00e9 spektrum mikroorganism\u016f a p\u0159edch\u00e1zet tvorb\u011b ne\u017e\u00e1douc\u00edch biofilm\u016f,\u201c<\/em> dod\u00e1v\u00e1 Petr Hauschwitz.<\/p>\n<p>V\u00fdsledky projektu znamenaj\u00ed v\u00fdznamn\u00fd p\u0159\u00ednos k sou\u010dasn\u00fdm poznatk\u016fm o efektivn\u00ed v\u00fdrob\u011b funk\u010dn\u00edch nanostruktur a budou publikov\u00e1ny v presti\u017en\u00edch odborn\u00fdch \u010dasopisech.<\/p>\n<h6>V\u011bdeck\u00e1 spolupr\u00e1ce p\u0159in\u00e1\u0161\u00ed praktick\u00e9 v\u00fdsledky<\/h6>\n<p>Interdisciplin\u00e1rn\u00ed spolupr\u00e1ce mezi mikrobiology z BIOCEV a laserov\u00fdmi specialisty z HiLASE ji\u017e p\u0159in\u00e1\u0161\u00ed prvn\u00ed hmatateln\u00e9 v\u00fdsledky. <em>\u201eNa\u0161e spole\u010dn\u00e9 \u00fasil\u00ed vedlo k vytvo\u0159en\u00ed prvn\u00edch typ\u016f povrchov\u00fdch \u00faprav, kter\u00e9 sni\u017euj\u00ed riziko mikrobi\u00e1ln\u00ed kontaminace,\u201c<\/em> \u0159\u00edk\u00e1 Petr Hauschwitz. \u201eTato \u00fasp\u011b\u0161n\u00e1 f\u00e1ze n\u00e1s utvrzuje v tom, \u017ee jdeme spr\u00e1vn\u00fdm sm\u011brem. Nyn\u00ed se zam\u011b\u0159\u00edme na dal\u0161\u00ed v\u00fdvoj topografi\u00ed, abychom roz\u0161\u00ed\u0159ili \u00fa\u010dinnost proti v\u00edce typ\u016fm mikrob\u016f,\u201c zd\u016fraz\u0148uje.<\/p>\n<p>Tyto \u00fapravy maj\u00ed potenci\u00e1l, aby zlep\u0161ily hygienick\u00e9 standardy ve zdravotnictv\u00ed. Technologie vyu\u017e\u00edvaj\u00edc\u00ed dynamick\u00e9 tvarov\u00e1n\u00ed laserov\u00e9ho svazku a interferen\u010dn\u00ed obr\u00e1b\u011bn\u00ed nejen\u017ee br\u00e1n\u00ed p\u0159ilnut\u00ed mikroorganism\u016f a n\u00e1sledn\u00e9 tvorb\u011b biofilm\u016f, ale tak\u00e9 v\u00fdrazn\u011b urychluj\u00ed a zlev\u0148uj\u00ed v\u00fdrobn\u00ed procesy.<\/p>\n<h6>St\u0159edo\u010desk\u00e9 Silicon Valley<\/h6>\n<p>Ob\u011b v\u011bdeck\u00e1 centra, BIOCEV a HiLASE, jsou sou\u010d\u00e1st\u00ed v\u011bdecko-technologick\u00e9ho klastru STAR (Science and Technology Advanced Region), kter\u00fd vznikl v roce 2015 nedaleko Prahy mezi obcemi Vestec, Zlatn\u00edky-Hodkovice a Doln\u00ed B\u0159e\u017eany. Na \u00fazem\u00ed o rozloze 23 km2 p\u016fsob\u00ed v\u00edce ne\u017e 1000 v\u011bdc\u016f a student\u016f v oblasti biotechnologi\u00ed, biomedic\u00edny, laser\u016f a laserov\u00fdch aplikac\u00ed.<\/p>\n<p>V\u00fdsledky b\u00e1d\u00e1n\u00ed v\u011bdc\u016f z BIOCEV sm\u011b\u0159uj\u00ed k n\u00e1vrh\u016fm nov\u00fdch l\u00e9k\u016f a l\u00e9\u010debn\u00fdch metod, nap\u0159. proti rakovin\u011b, neplodnosti nebo covid-19. Fyzici z Doln\u00edch B\u0159e\u017ean vyv\u00edjej\u00ed supermodern\u00ed lasery, kter\u00e9 se uplat\u0148uj\u00ed v pr\u016fmyslov\u00fdch aplikac\u00edch, medic\u00edn\u011b, ve vesm\u00edru i obran\u011b.<\/p>\n<p><em>\u201eNa\u0161\u00edm c\u00edlem je maxim\u00e1ln\u011b vyu\u017e\u00edt v\u011bdeck\u00e9ho a technologick\u00e9ho potenci\u00e1lu, kter\u00fd p\u0159edstavuj\u00ed v\u00fdzkumn\u00e9 organizace a firmy ve STAR. Po\u0159\u00e1d\u00e1me networkingov\u00e9 a vzd\u011bl\u00e1vac\u00ed akce, kde spole\u010dn\u011b diskutujeme o mo\u017enostech podpory v\u011bdc\u016f i mezioborov\u00e9 spolupr\u00e1ce. V\u00fdsledky dal\u0161\u00edch v\u011bdeck\u00fdch synergi\u00ed nap\u0159\u00ed\u010d p\u0159\u00edrodn\u00edmi a technick\u00fdmi obory mohou b\u00fdt velk\u00fdm p\u0159\u00ednosem pro spole\u010dnost,\u201c<\/em> \u0159\u00edk\u00e1 Jana De Merlier, \u0159editelka STAR klastru.<\/p>\n<h6>Interdisciplin\u00e1rn\u00ed projekt LasApp: spojen\u00ed laser\u016f s mikrobiologi\u00ed<\/h6>\n<p>Projekt LasApp rozv\u00edj\u00ed centra v\u011bdeck\u00e9 excelence a kompetence v laserov\u00e9 technice se zam\u011b\u0159en\u00edm na vl\u00e1knov\u00e9 a tenkodiskov\u00e9 lasery a jejich potenci\u00e1ln\u00ed aplikace pro chytrou v\u00fdrobu, vesm\u00edrn\u00e9 a biotechnologick\u00e9 aplikace. Spojuje excelentn\u00ed laserov\u00e1 v\u00fdzkumn\u00e1 centra Akademie v\u011bd \u010cesk\u00e9 republiky s dal\u0161\u00edmi \u0161pi\u010dkov\u00fdmi pracovi\u0161ti. V tomto p\u0159\u00edpad\u011b experti z projektu LasApp a v\u011bdecko-technologick\u00e9ho klastru STAR v r\u00e1mci siln\u011b interdisciplin\u00e1rn\u00edho p\u0159\u00edstupu kombinuj\u00ed \u0161pi\u010dkov\u00e9 laserov\u00e9 technologie s unik\u00e1tn\u00edm know-how v oblasti biofilm\u016f. V\u00fdzkumn\u00edci se mohou op\u0159\u00edt o robustn\u00ed z\u00e1zem\u00ed pro pr\u00e1ci s daty, automatizaci, robotizaci a vyu\u017eit\u00ed um\u011bl\u00e9 inteligence.
