Pracovn\u00edci z Mikrobiologick\u00e9ho \u00fastavu AV \u010cR nyn\u00ed v mezin\u00e1rodn\u00ed spolupr\u00e1ci s Tokijskou univerzitou vy\u0159e\u0161ili strukturu kl\u00ed\u010dov\u00e9ho kondenza\u010dn\u00edho (spojovac\u00edho) enzymu, kter\u00fd se vyu\u017e\u00edv\u00e1 pro biosynt\u00e9zu antibiotik, a objasnili na molekul\u00e1rn\u00ed \u00farovni, jak funguje. Svou strukturou se vymyk\u00e1 jak\u00e9mukoli zn\u00e1m\u00e9mu proteinu. Na molekul\u00e1rn\u00ed \u00farovni jej lze p\u0159irovnat k ptakopyskovi, kter\u00fd takt\u00e9\u017e zd\u00e1nliv\u011b poch\u00e1z\u00ed z r\u016fzn\u00fdch zv\u00ed\u0159at.<\/p>\n<h6>L\u00e9ta v\u00fdzkumu funk\u010dn\u00edch antibiotik<\/h6>\n<p>L\u00e1tky \u00fa\u010dinn\u00e9 i proti ob\u00e1van\u00fdm zlat\u00fdm stafylokok\u016fm a jin\u00fdm nebezpe\u010dn\u00fdm bakteri\u00edm p\u0159ipravuj\u00ed v\u011bdci z Mikrobiologick\u00e9ho \u00fastavu AV \u010cR ve spolupr\u00e1ci s malou \u010deskou firmou Santiago Chemicals u\u017e n\u011bkolik let. Vych\u00e1zej\u00ed z v\u00fdzkumu, kter\u00fd za\u010dal pod veden\u00edm Ji\u0159\u00edho Janaty v roce 2017. Tehdy v\u011bdci p\u0159i\u0161li s nov\u00fdm, \u00fa\u010dinn\u011bj\u0161\u00edm antibiotikem CELIN, kter\u00e9 kombinuje stavebn\u00ed kameny dvou p\u0159\u00edrodn\u00edch l\u00e1tek \u2013 CELesticetinu a LINkomycinu. Od t\u00e9 doby spole\u010dn\u011b p\u0159ipravili des\u00edtky odvozen\u00fdch l\u00e1tek, n\u011bkolik je \u00fa\u010dinn\u00fdch i proti bakteri\u00edm rezistentn\u00edm k antibiotik\u016fm.<\/p>\n<p>V sou\u010dasnosti v r\u00e1mci projektu N\u00e1rodn\u00ed centrum virologie a bakteriologie u\u010dinili dal\u0161\u00ed p\u0159elomov\u00fd krok, kdy\u017e s t\u00fdmem profesora Ikura Abe z Tokijsk\u00e9 univerzity objasnili strukturu podivn\u00e9ho enzymu. Ten je z\u00e1sadn\u00ed pro p\u0159\u00edpravu zmi\u0148ovan\u00e9ho celesticetinu, bakteriostatick\u00e9ho antibiotika.<\/p>\n<p><em>\u201eV letech 2015\u20132018 jsme v\u00fdraznou m\u011brou p\u0159isp\u011bli k vy\u0159e\u0161en\u00ed podivuhodn\u00e9 biosynt\u00e9zy linkosamidov\u00fdch antibiotik (celesticetin a linkomycin), v nich\u017e se jednotliv\u00e9 stavebn\u00ed kameny sestavuj\u00ed jako v molekul\u00e1rn\u00ed verzi stavebnice Lego. Kdy\u017e jsme si tyto principy vyzkou\u0161eli p\u0159i konstrukci hybridn\u00ed l\u00e1tky CELIN, napadlo n\u00e1s, \u017ee by na\u0161e stavebn\u00ed mo\u017enosti byly v\u00fdrazn\u011b \u0161ir\u0161\u00ed, kdybychom rozum\u011bli funkci z\u00e1sadn\u00edch enzym\u016f z biosynt\u00e9zy p\u0159\u00edrodn\u00edch linkosamid\u016f,\u201c<\/em> vysv\u011btluje Ji\u0159\u00ed Janata z Mikrobiologick\u00e9ho \u00fastavu AV \u010cR.<\/p>\n<h6>V\u00fdjime\u010dn\u00e9 v\u00fdsledky d\u00edky \u010desko-japonsk\u00e9 spolupr\u00e1ci<\/h6>\n<p>U\u017e podle sekvence zkouman\u00fdch enzym\u016f (po\u0159ad\u00ed aminokyselin v nich) bylo jasn\u00e9, \u017ee budou hodn\u011b odli\u0161n\u00e9 od v\u0161ech dosud zn\u00e1m\u00fdch protein\u016f. <em>\u201eM\u011b fascinoval kl\u00ed\u010dov\u00fd kondenza\u010dn\u00ed enzym, nepodobal se v\u016fbec ni\u010demu. Intuice v biologii \u0159\u00edk\u00e1, \u017ee pr\u00e1v\u011b unik\u00e1tn\u00ed syst\u00e9my p\u0159in\u00e1\u0161ej\u00ed ta nejz\u00e1sadn\u011bj\u0161\u00ed p\u0159ekvapen\u00ed a p\u0159elomov\u00e9 objevy. Oslovil jsem tehdy profesora Abe, kter\u00fd se zab\u00fdv\u00e1 pr\u00e1v\u011b takov\u00fdmi neobvykl\u00fdmi enzymy tzv. speci\u00e1ln\u00edho (sekund\u00e1rn\u00edho) metabolismu p\u0159\u00edrodn\u00edch l\u00e1tek, domluvili spolupr\u00e1ci p\u0159i \u0159e\u0161en\u00ed struktury a funkce pr\u00e1v\u011b publikovan\u00e9ho kondenza\u010dn\u00edho enzymu,\u201c<\/em> popisuje vznik spolupr\u00e1ce s Japonci Ji\u0159\u00ed Janata.<\/p>\n<p>Do t\u011bch nejpresti\u017en\u011bj\u0161\u00edch \u010dasopis\u016f \u0159ady Nature se \u010de\u0161t\u00ed v\u011bdci na \u00farovni rovnocenn\u00fdch partner\u016f prosad\u00ed zhruba jednou a\u017e dvakr\u00e1t za rok. To jen potvrzuje, \u017ee v\u00fdsledky jsou v\u00fdjime\u010dn\u00e9. Spolupr\u00e1ce s japonskou skupinou se postupn\u011b roz\u0161\u00ed\u0159ila o dal\u0161\u00ed t\u0159i proteiny kl\u00ed\u010dov\u00e9 pro skl\u00e1d\u00e1n\u00ed a opracov\u00e1n\u00ed stavebn\u00edch kamen\u016f antibiotik. Strukturu a funkce jednoho z nich objas\u0148uje druh\u00fd spole\u010dn\u00fd \u010dl\u00e1nek v \u010dervencov\u00e9m \u010d\u00edsle jin\u00e9ho exkluzivn\u00edho \u010dasopisu Angewandte Chemie. Dal\u0161\u00ed dva \u010dl\u00e1nky ob\u011b skupiny p\u0159ipravuj\u00ed.<\/p>\n<h6>N\u00e1ro\u010dn\u00e1 a nejist\u00e1 cesta k \u00fasp\u011bchu<\/h6>\n<p>Strukturu z\u00e1kladn\u00ed formy kondenza\u010dn\u00edho enzymu vy\u0159e\u0161il Stanislav Kadl\u010d\u00edk z Mikrobiologick\u00e9ho \u00fastavu AV \u010cR, kter\u00fd v roce 2019 pob\u00fdval jako postdoktorand v tokijsk\u00e9 skupin\u011b.<\/p>\n<p><em>\u201eStudiu struktury proteinu p\u0159edch\u00e1z\u00ed jeho \u010di\u0161t\u011bn\u00ed a krystalizace, co\u017e je dlouh\u00e1 a nevyzpytateln\u00e1 pr\u00e1ce bez jistoty \u00fasp\u011bchu. Ka\u017ed\u00fd protein vy\u017eaduje specifick\u00e9 podm\u00ednky. Kdy\u017e m\u00e1te \u0161t\u011bst\u00ed a poda\u0159\u00ed se z\u00edskat krystal proteinu v kvalit\u011b dostate\u010dn\u00e9 pro m\u011b\u0159en\u00ed struktury, nen\u00ed je\u0161t\u011b vyhr\u00e1no a jste tak ve t\u0159etin\u011b cesty k c\u00edli. K vy\u0159e\u0161en\u00ed mechanismu funkce neobvykl\u00e9ho proteinu mus\u00edte m\u00edt i krystaly se substr\u00e1ty enzymu, \u010dasto pr\u00e1v\u011b tak neobvykl\u00fdmi a tedy nedostupn\u00fdmi. A pak se mus\u00ed p\u0159ipravit a otestovat upraven\u00e9 formy enzymu. Takov\u00fd v\u00fdzkum trv\u00e1 i n\u011bkolik let a kdykoli m\u016f\u017eete narazit,\u201c<\/em> vysv\u011btluje n\u00e1ro\u010dnou pr\u00e1ci Stanislav Kadl\u010d\u00edk.<\/p>\n<p>Dal\u0161\u00ed spoluautor \u010dl\u00e1nku Zden\u011bk Kamen\u00edk z Mikrobiologick\u00e9ho \u00fastavu AV \u010cR k tomu dod\u00e1v\u00e1: <em>\u201eNejde p\u0159itom jen o p\u0159ek\u00e1\u017eky, kter\u00e9 n\u00e1m stav\u00ed p\u0159\u00edroda. Kdy\u017e se dnes mluv\u00ed o \u0161krtech ve v\u00fddaj\u00edch na v\u011bdu, m\u00e1lokdo si uv\u011bdomuje, jak je takov\u00e1 dlouhodob\u00e1 mezin\u00e1rodn\u00ed spolupr\u00e1ce k\u0159ehk\u00e1. To, co se buduje a do \u010deho se investuje n\u011bkolik let, se d\u00e1 i za jedin\u00fd rok zni\u010dit. Partne\u0159i mus\u00ed pokra\u010dovat v \u0159e\u0161en\u00ed sv\u00fdch projekt\u016f a v\u00fdchov\u011b student\u016f bez ohledu na to, jak\u00e1 je situace u n\u00e1s. A je jasn\u00e9, \u017ee pr\u00e1v\u011b na kvalit\u011b dne\u0161n\u00edch v\u011bdeck\u00fdch v\u00fdsledk\u016f stoj\u00ed na\u0161e konkurenceschopnost v p\u0159\u00ed\u0161t\u00edch letech.\u201c<\/em><\/p>\n<h6>Porozum\u011bn\u00ed \u0159e\u010di mikroorganism\u016f je cestou ke zcela nov\u00fdm zp\u016fsob\u016fm l\u00e9\u010dby infekc\u00ed<\/h6>\n<p>P\u0159irovn\u00e1n\u00ed v\u00fdstavby antibiotika z jednotliv\u00fdch stavebn\u00edch d\u00edl\u016f ke skl\u00e1da\u010dce Lega nen\u00ed n\u00e1hodn\u00e9. Antibiotika jsou speci\u00e1ln\u00edmi molekul\u00e1rn\u00edmi stavbami na m\u00edru, p\u0159esn\u011b zapadaj\u00ed do z\u00e1sadn\u00edch biologick\u00fdch struktur mikroorganism\u016f. Proto jsou tak \u00fa\u010dinn\u00e1 jen proti bakteri\u00edm a pro \u010dlov\u011bka jsou ne\u0161kodn\u00e1. Rezistence k antibiotik\u016fm n\u011bjak\u00fdm zp\u016fsobem tuto do sebe zapadaj\u00edc\u00ed skl\u00e1da\u010dku ru\u0161\u00ed a antibiotika p\u0159est\u00e1vaj\u00ed fungovat.<\/p>\n<p>Gabriela Bal\u00edkov\u00e1 Novotn\u00e1, odbornice na antibiotickou rezistenci z t\u00fdmu J. Janaty, dopl\u0148uje: <em>\u201eV p\u0159\u00edrod\u011b vedle sebe po miliony let existuj\u00ed antibiotika i rezistence, a p\u0159esto antibiotika p\u0159etrv\u00e1vaj\u00ed. Antibiotika a rezistence jsou toti\u017e dv\u011bma ned\u00edln\u00fdmi sou\u010d\u00e1stmi \u0161ir\u0161\u00edho syst\u00e9mu chemick\u00e9 komunikace mikroorganism\u016f, zjednodu\u0161en\u011b \u0159e\u010deno bakteri\u00ed. Kdy\u017e z p\u0159\u00edrody odpozorujeme tyto principy, \u201aporozum\u00edme \u0159e\u010di bakteri\u00ed\u2018, m\u016f\u017eeme v budoucnu l\u00e9\u010dit infek\u010dn\u00ed nemoci zcela nov\u00fdm zp\u016fsobem. Nebude nap\u0159\u00edklad nutn\u00e9 infek\u010dn\u00ed agens antibiotiky zab\u00edt. Nam\u00edsto toho bude mo\u017en\u00e9 patogenn\u00edm bakteri\u00edm vyslat sign\u00e1ly, kter\u00e9 je p\u0159esv\u011bd\u010d\u00ed k \u00fastupu. Bude to takov\u00e1 informa\u010dn\u00ed v\u00e1lka nam\u00edsto t\u00e9 likvida\u010dn\u00ed. To, si mysl\u00edm, je cesta pro p\u0159\u00ed\u0161t\u00ed desetilet\u00ed.\u201c<\/em><\/p>\n<p><em>\u201ePorozum\u011bn\u00ed a vyu\u017eit\u00ed \u0159e\u010di mikroorganism\u016f je \u00fast\u0159edn\u00edm konceptem nejen t\u00fdmu Ji\u0159\u00edho Janaty, ale i sjednocuj\u00edc\u00ed viz\u00ed na\u0161eho \u00fastavu pro p\u0159\u00ed\u0161t\u00ed obdob\u00ed. Je to t\u00e9ma p\u011btilet\u00e9ho projektu podan\u00e9ho spole\u010dn\u011b s V\u0160CHT a UK v r\u00e1mci v\u00fdzvy \u0160pi\u010dkov\u00fd v\u00fdzkum OP JAK.
